Growth kinetics, effect of carbon substrate in biosynthesis of mcl-PHA by Pseudomonas putida Bet001 BJM

Growth associated biosynthesis of medium chain length poly-3-hydroxyalkanoates (mcl-PHA) in Pseudomonas putida Bet001 isolated from palm oil mill effluent was studied. Models with substrate inhibition terms described well the kinetics of its growth. Selected fatty acids (C8:0 to C18:1) and ammonium were used as carbon and nitrogen sources during growth and PHA biosynthesis, resulting in PHA accumulation of about 50 to 69% (w/w) and PHA yields ranging from 10.12 g L-1 to 15.45 g L-1, respectively. The monomer composition of the PHA ranges from C4 to C14, and was strongly influenced by the type of carbon substrate fed. Microbiological processes were used for chitin and chitosan productions by Cunninghamella elegans (UCP 542) grown in a new economic culture medium. The assay was carried out to evaluate the growth of C. elegans using yam bean (Pachyrhizus erosus L. Urban) medium, in different times of growth (24, 48, 72 and 96 hrs), incubated at 28ºC in an orbital shaker at 150 rpm. The lyophilized biomass was determined by gravimetry. The polysaccharides were extracted by alkali-acid treatment, and characterized by infrared spectroscopy, titration and viscosity. C. elegans grown in the yam bean medium and produced higher yields of biomass (24.3 g/ mL) in 96 hrs. It is well known that microorganisms well-adapted to survival in extreme ecosystems could be considered as new sources of biomolecules that have biotechnological importance. On French Polynesian atolls, microbial mats are developing in water ponds exposed to fluctuations in physical and chemical parameters. In these microbial mats, which are called "kopara" by the inhabitants, bacteria coexist with cyanobacteria, and a synergistic relationship may exist between these two types of living microorganisms. Se elaboraron películas a partir de disoluciones a 3 % (m/v) de quitosana en ácido láctico a 1% (v/v), empleando glicerina, sorbitol o polietilenglicol como plastificante en concentraciones de 0,25 a 0,50 mL/g de quitosana. Se midieron el esfuerzo tensil, elongación, fuerza de ruptura, deformación y el espesor de las películas, con el objetivo de evaluar la influencia de la masa molar y concentración de plastificante sobre sus propiedades mecánicas. This study describes the techniques used for the identification of a novel Vibrio bacterium and preliminary characterization of its EPS. ABSTRACT This paper describes the preparation and characterization of alginate beads coated with gelatin and containing Lactobacillus rhamnosus. Capsules were obtained by extrusion method using CaCl2 as cross linker. An experimental design was performed using alginate and gelatin concentrations as the variables investigated, while the response variable was the concentration of viable cells. Beads were characterized in terms of size, morphology, scanning electron microscopy (SEM), moisture content, Fourier Transform Infrared Spectrometry (FTIR), thermal behavior and cell viability during storage. Production of biopolymers by Pseudomonas aeruginosa isolated from marine source BABT

Two bacterial strains, Pseudomonas aeruginosa CMG607w and CMG1421 produce commercially important biopolymers. CMG607w isolated from the sediments of Lyari outfall to Arabian Sea synthesize the mcl-polyhydroxyalkanoates from various carbon sources. The production of PHAs was directly proportional to the incubation periods. Other strain CMG1421, a dry soil isolate, produced high viscous water absorbing extracellular acidic polysaccharide when it was grown aerobically in the minimal medium containing glucose or fructose or sucrose as sole source of carbon. Properties of films obtained from biopolymers of different origins for skin lesions therapy BABT

In this study, the effects of the origin of xanthan used, in combination with chitosan, to prepare films for the treatment of skin lesions were evaluated. The characteristics of the films obtained with xanthan commercially available for the food industry sector and xanthan originated from a fermentation process conducted in a pilot plant were compared. Results showed that the source did not strongly interfere in many of the properties of the films, such as the mechanical properties, cytotoxicity to L929 cells, absorption of simulated body fluid and culture medium, stability in water and saline solution. The distribution of selected aromatic compounds and microbiology were assessed in superficial sediments from Suruí Mangrove, Guanabara Bay. Samples were collected at 23 stations, and particle size, organic matter, aromatic compounds, microbiology activity, biopolymers, and topography were determined. The concentration of aromatic compounds was distributed in patches over the entire mangrove, and their highest total concentration was determinated in the mangrove's central area. Particle size differed from most mangroves in that Suruí Mangrove has chernies on the edges and in front of the mangrove, and sand across the whole surface, which hampers the relationship between particle size and hydrocarbons. Polyhydroxyalkanoates (PHAs) can be produced by microorganisms and are a biodegradable alternative to fossil-fuel based plastics. Currently, the focus is on reducing production costs by exploring alternative substrates for PHAs production, and on producing copolymers which are less brittle than monomers. Accordingly, this study used a substrate consisting of wastewater from waste-glycerol fermentation, supplemented with different amounts of acetic and propionic acids.
